Lamivudine - Drug Information
Typical usage: :Portal systemic encephalopathy, chronic or habitual constipation.
Probable Side Effects: :The symptomatic adverse reactions produced by Lamivudine are more or less tolerable and if they become severe, they can be treated symptomatically, these include Headache, Fatigue, Diarrhea, Abdominal pain, Malaise, Nausea and vomiting, Respiratory tract infections, Abdominal discomfort.
Drug Interaction: Trimethoprim/sulfamethoxazole (Bactrim) increases the concentration of lamivudine in the body.
Mechanism of Action: Lamivudine is a synthetic nucleoside analogue and is phosphorylated intracellularly to its active 5'-triphosphate metabolite, lamivudine triphosphate (L-TP). This nucleoside analogue is incorporated into viral DNA by HIV reverse transcriptase and HBV polymerase, resulting in DNA chain termination.
Pregnancy Impact: Animal reproduction studies have shown an adverse effect on the fetus and there are no adequate and well-controlled studies in humans, but potential benefits may warrant use of the drug in pregnant women despite potential risks.
Impact During Lactation: There is positive evidence of risk to a breastfed infant or to breastmilk production, but the benefits of use in breastfeeding mothers may be acceptable despite the risk to the infant (e.g. if the drug is needed in a life-threatening situation or for a serious disease for which safer drugs cannot be used or are ineffective).
Stavudine - Drug Information
Typical usage: :HIV infection.
Probable Side Effects: :Flatulence, Dizziness, Headache, Drowsiness, Fatigue, Anorexia, Diarrhea, Fever, Myalgia, Anxiety, Dyspnea, Rashes, Urticaria, Depression, Cough, Arthralgia, Nausea and vomiting, GI disturbance, Abnormal dreams.
Drug Interaction: Zidovudine prevents the conversion of stavudine to its active form (stavudine triphosphate) and this reduces the action of stavudine against the HIV virus. Therefore stavudine should not be combined with zidovudine.
Mechanism of Action: Stavudine inhibits the activity of HIV-1 reverse transcriptase (RT) both by competing with the natural substrate dGTP and by its incorporation into viral DNA.
Pregnancy Impact: There is positive evidence of human fetal risk based on adverse reaction data from investigational or marketing experience or studies in humans, but potential benefits may warrant use of the drug in pregnant women despite potential risks.
Impact During Lactation: Studies in breastfeeding mothers have demonstrated that there is significant and documented risk to the infant based on human experience, or it is a medication that has a high risk of causing significant damage to an infant. The risk of using the drug in breastfeeding women clearly outweighs any possible benefit from breastfeeding. The drug is contraindicated in women who are breastfeeding an infant.
