Domperidone - Drug Information
Typical usage: :Acute nausea and vomiting, dyspepsia, functional dyspepsia, gastric distension pain due to pressure on the stomach, gastro-oesophageal reflux disease, nausea and vomiting, nausea and vomiting (chemotherapy induced), nausea and vomiting (L-dopa induced).
Probable Side Effects: :Tardive dyskinesia, parkinsonism, extrapyramidal dystonic reactions, convulsions, coma, GI disturbances, extrapyramidal effects, thirst, headache, drowsiness, diarrhea, anxiety, restlessness, insomnia, nervousness, skin rashes, dry mouth, itching, galactorrhea, breast enlargement, soreness, hyperprolactinemia.
Drug Interaction: Domperidone is known to interact with other drugs like apomorphine, bromocriptine (Mesylate), cabergoline, cinnarizine, lisuride, oxycodone, pergolide, piribedil. Always consult your physician for the change of dose regimen or an alternative drug of choice that may strictly be required.
Mechanism of Action: Domperidone facilitates gastric emptying and decreases small bowel transit time by increasing esophageal and gastric peristalsis and by lowering esophageal sphincter pressure. Antiemetic: The antiemetic properties of domperidone are related to its dopamine receptor blocking activity at both the chemoreceptor trigger zone and at the gastric level. It has strong affinities for the D2 and D3 dopamine receptors, which are found in the chemoreceptor trigger zone, located just outside the blood brain barrier, which - among others - regulates nausea and vomiting.
Pregnancy Impact: Animal reproduction studies have shown an adverse effect on the fetus and there are no adequate and well-controlled studies in humans, but potential benefits may warrant use of the drug in pregnant women despite potential risks.
Impact During Lactation: There are no controlled studies in breastfeeding women, however the risk of untoward effects to a breastfed infant is possible; or, controlled studies show only minimal non-threatening adverse effects. Drugs should be given only if the potential benefit justifies the potential risk to the infant.
Esomeprazole - Drug Information
Typical usage: :Gastric ulcer, gastro-esophageal reflux disease, zollinger-ellison syndrome.
Probable Side Effects: :Flatulence, dizziness, headache, diarrhea, constipation, abdominal pain, dry mouth, blurred vision, rashes, pruritus, malaise, arthralgia, nausea and vomiting, musculoskeletal pain.
Drug Interaction: Esomeprazole is known to interact with other drugs like disulfiram, phenytoin (Na), warfarin (Na). Always consult your physician for the change of dose regimen or an alternative drug of choice that may strictly be required.
Mechanism of Action: Esomeprazole is a proton pump inhibitor that suppresses gastric acid secretion by specific inhibition of the H+/K+-ATPase in the gastric parietal cell. By acting specifically on the proton pump, Esomeprazole blocks the final step in acid production, thus reducing gastric acidity.
Pregnancy Impact: Animal reproduction studies have failed to demonstrate a risk to the fetus and there are no adequate and well-controlled studies in pregnant women.
Impact During Lactation: Studies in breastfeeding mothers have demonstrated that there is significant and documented risk to the infant based on human experience, or it is a medication that has a high risk of causing significant damage to an infant. The risk of using the drug in breastfeeding women clearly outweighs any possible benefit from breastfeeding. The drug is contraindicated in women who are breastfeeding an infant.
