Lafutidine - Drug Information
Typical usage: :Used for treatment of ulcer, suppresses gastric acid secretion.
Probable Side Effects: :Constipation, hyperuricaemia, increase in LFT and total bilirubin, gynaecomastia, anorexia, hallucinations, increase in liver enzymes, increased level of blood urea, loss of appetite.

Mechanism of Action: Acid suppressive action: Lafutidine is absorbed in the small intestine, reaches gastric cells via the systemic circulation, and then directly and rapidly binds to gastric cell histamine H2 receptors, thereby inhibiting the stimulation of cAMP and a resultant decrease in acid production (antisecretory action). It causes a sustained increase in intracellular Ca2+ ion concentration in endothelial cells resulting in the release of calcitonin gene related peptide (CGRP), which causes acid suppression by decreasing the vagal tone. Gastroprotective action: Lafutidine induced CGRP release stimulates nitric oxide (NO) production in endothelial cells, where NO participates in the regulation of gastric mucosal blood flow through vasodilation in the gastric microvasculature and increases mucin biosynthesis resulting in gastroprotection. Lafutidine inhibits secretion of interleukin-8, which in turn inhibits sequestration of neutrophils at the site of inflammation, thereby preventing mucosal inflammation. It also inhibits neutrophil activation which reduces damage caused by free radicals, thereby reducing inflammation.
Pregnancy Impact: Animal reproduction studies have shown an adverse effect on the fetus and there are no adequate and well-controlled studies in humans, but potential benefits may warrant use of the drug in pregnant women despite potential risks.
Impact During Lactation: There is positive evidence of risk to a breastfed infant or to breastmilk production, but the benefits of use in breastfeeding mothers may be acceptable despite the risk to the infant (e.g. if the drug is needed in a life-threatening situation or for a serious disease for which safer drugs cannot be used or are ineffective).
